PARP plays a key role in the repair of ssDNA
breaks via base excision repair (BER) pathway

• Binds directly to sites of DNA damage
Nicotinamide + • Once activated, uses NAD as a substrate

to add large, branched chains of
pADPr polymers (ie, PARylation)
• Recruits other DNA repair enzymes
to site of damage
